How to seal a pcr plate​?

Achieving a secure, contamination-free seal for your PCR or qPCR plates is critical for accurate results. Whether using aluminum sealing film for PCR or adhesive seals, the right materials and tools—like a qPCR plate sealer or PCR plate sealer tool—ensure optimal performance in high-throughput labs.

Start by selecting a high-quality aluminum sealing film for PCR, designed to withstand extreme temperatures (up to 100°C+) and prevent evaporation during thermal cycling. Its durable, heat-resistant properties make it ideal for qPCR applications requiring precise temperature control. For adhesive-based sealing, opt for non-sticky, optically clear films that eliminate autofluorescence and ensure compatibility with fluorescence detection systems.

Using a specialized PCR plate sealer tool streamlines the process, applying uniform pressure to create an airtight seal across all wells. This minimizes the risk of cross-contamination and sample loss, especially when working with sensitive qPCR assays. For automated workflows, a qPCR plate sealer with adjustable settings ensures consistent sealing force, enhancing reproducibility in high-stakes experiments.

Key considerations when sealing plates:

To remove a qPCR plate sealer, peel it back slowly from one corner at a low angle, rather than pulling straight up. For heat-sealed films, use the appropriate sealer tool to cut or peel it off carefully.

In addition to 96-well and 384-well PCR plates, sealing films are also suitable for ELISA plates and deep-well plates, meeting various experimental needs.

Optical pcr film are recommended to ensure that fluorescent signals can pass through the film without affecting the test results.

PCR films are disposable. To avoid cross-contamination and experimental errors, reuse is not recommended.

  • For routine PCR amplification, self-adhesive pcr plate seals or pressure-sensitive pcr sealing film can be used. They are easy to use and meet the requirements for preventing evaporation and cross-contamination.
  • If the experiment involves long cycles or high temperatures, heat-sealing films are recommended. They provide a stronger seal and are less likely to warp at high temperatures, making them suitable for large-scale and high-stability experiments.
  • If real-time quantitative PCR (qPCR) or fluorescence detection is required, optically transparent sealing films should be selected to ensure that the fluorescent signal accurately transmits through the film and avoid affecting detection sensitivity.
