How to autoclave microcentrifuge tubes?
- High-pressure steam sterilization (preferred method)
As the most commonly used sterilization method in laboratories, high-pressure steam sterilization achieves thorough sterilization through high-temperature and high-pressure saturated steam. The parameters need to be set to 121°C and 15 pounds per square inch (psi) pressure for 15-20 minutes. Before loading, make sure that the caps of all centrifuge tubes are completely closed, and use a special sterilization box to place the micro centrifuge tubes vertically to avoid deformation of the tubes caused by stacking. It is recommended that the single sterilization volume does not exceed 80% of the volume of the autoclave to ensure sufficient penetration of the steam cycle. This method can effectively inactivate various microorganisms including bacterial spores, and is particularly suitable for the sterilization needs of conventional laboratory consumables. It should be noted that although the product has been verified by 100 repeated sterilizations, in order to ensure the sealing of the buckle structure and the mechanical strength of the tube body, it is recommended that the cumulative sterilization times of a single tube do not exceed 50 times.
- Gamma ray irradiation sterilization method (for pre-sterilized microfuge tube only)
For factory-presterilized individual packaged products, gamma ray sterilization provides a solution that does not require high-temperature treatment. Using an irradiation dose of 25-40 kGy, this technology meets the sterilization standards of medical devices and can penetrate the packaging materials at room temperature to achieve all-round sterilization. It is especially suitable for precision experimental scenarios that are sensitive to temperature, such as single-cell sequencing and sterile cell culture. The sterilized product maintains the convenience of being ready to use, and the tube body will not be deformed due to thermal effects, and the original physical properties are completely retained. This method must be completed in a professional irradiation center. Laboratories usually directly purchase pre-sterilized packaging products to avoid the risk of cross-contamination.
How to balance 5 tubes in a centrifuge?
- Basic balancing principle
Symmetrical balance: The centrifuge rotor must maintain a symmetrical mass distribution, that is, the total weight of the centrifuge tubes in the symmetrical position is equal.
Error range: When centrifuging at high speed, the weight difference in the symmetrical position should be ≤0.1g; the error range can be appropriately relaxed for low-speed centrifugation (it is recommended to be as accurate as possible)

Does the EP tube cap seal well? Will it leak?
High-quality EP tubes caps are designed to be tight and provide a good seal, preventing sample evaporation or leakage. However, during high-temperature sterilization or extremely high centrifugal forces, it is recommended not to overfill the tube with sample to prevent excessive pressure that could cause the cap to open.
