FAQ Of ELISA Plate 96 Well
Is there a difference between a 96 well plate ELISA and a regular 96-well plate?
Yes. 96 well ELISA plates typically have a special surface treatment to enhance protein or antibody adsorption. However, regular 96-well plates (such as cell culture plates) have different surface properties and are not suitable for ELISA testing.
How should I choose high binding ELISA plates or medium binding ELISA plates format?
The protein binding capacity of microplates is generally classified into three levels:
- Low Binding: Preferentially binds molecules with strong hydrophobic characteristics, such as lipids, lipoproteins, and large proteins. It is suitable for applications where non-specific adsorption needs to be minimized.
- Medium Binding: Suitable for biomolecules with both hydrophilic and hydrophobic properties, such as medium to large proteins, immunoglobulins, and albumin. It provides a balance between binding efficiency and background noise.
- High Binding: Offers strong surface adsorption capability and can bind a wide range of biomolecules with hydrophilic/hydrophobic properties, including small to large proteins, immunoglobulins, albumin, LPS, and other proteins. It is suitable for high-sensitivity assays.
How to choose the right ELISA microtiter plate?
- Color
Clear Plate: Suitable for absorbance-based assays, including quantitative and qualitative solid-phase immunoassays (ELISA) and binding assays. It allows direct optical reading.
White Plate: Ideal for luminescence-based assays such as bioluminescence and chemiluminescence. It enhances signal reflection and improves detection sensitivity.

- Binding capacity
High binding capacity: After surface treatment, the protein binding capacity is greatly enhanced, up to 300~400ng IgG/cm2, and the molecular weight of the main bound protein is >10kD. using this type of enzyme labeling plate can improve the sensitivity, and can relatively reduce the concentration and dosage of the encapsulated proteins, but the disadvantage is that it is easy to produce non-specific reaction. After antigen or antibody encapsulation, non-ionic detergents cannot effectively seal the site of unbound protein, and proteins need to be used as sealing agents.
Medium Binding: Passively binds to proteins via surface hydrophobic bonding and is suitable as a solid-phase carrier for large proteins with molecular weights >20kD, with a protein binding capacity of 200~300ng IgG/cm2. Due to its characteristic of binding only to large molecules, this type of plate is suitable for use as a solid-phase carrier for unpurified antibodies or antigens to minimize the potential for non-specific cross-reactivity. The plates can be used with inert proteins or non-ionic detergents as a sealing solution.

Difference between ELISA plates and cell culture plates
