What is the difference between screw-cap and snap-cap tubes?
Screw-cap tubes provide stronger sealing and are ideal for transportation and long-term storage, while snap-cap tubes allow faster opening and closing for routine laboratory use.
How to choose centrifuge tubes?
- Capacity: Small sample volumes are typically handled in 0.5 mL to 2 mL micro centrifuge tubes, while larger volumes are best handled in 15 ml centrifuge tubes or centrifuge for 50ml tubes.
- Bottoms types:
- Conical bottom centrifuge tubes: the sample is concentrated at the bottom, the reaction is mixed evenly, and it is suitable for micro-reaction systems.
- Round bottom centrifuge tubes: It matches the groove design of the PCR instrument heat block more closely, and the heat transfer is better; it reduces liquid residue.
- Self standing centrifuge tubes: It can stand directly on the workbench or ice box without the need for an additional tube rack, reducing the number of experimental steps.
- Material: Common polypropylene (PP) offers excellent chemical and temperature resistance, making it suitable for most molecular biology and chemistry experiments. However, if samples require resistance to organic solvents or require operation at higher temperatures, specialized tube materials are recommended.
- Centrifugal Force Resistance: Different grades of centrifuge tubes can withstand different maximum relative centrifugal forces (RCF), which should be adjusted according to the centrifuge speed.
- Caps: Common types include screw caps and snap caps. Screw caps offer a good seal and are suitable for long-term storage or experiments that need to prevent evaporation; snap caps are more convenient for quick operation.
- In addition, some experiments have strict requirements for sterility and the absence of enzyme contamination, such as molecular cloning or RNA experiments. Sterile centrifuge tubes that have been treated to be free of DNA and RNA enzymes should be selected. For light-sensitive samples, brown centrifuge tubes can be used.
Which tubes need to be centrifuged?
Not every laboratory tube needs centrifugation — it depends on the experiment type and sample handling requirements. Tubes that typically require centrifugation include:
- Blood collection tubes: After drawing blood, centrifugation is needed to separate plasma or serum from cells. For example, serum tubes (with clot activator) and plasma tubes (with anticoagulant like EDTA or heparin).
- Cell culture tubes or conical centrifuge tubes: When harvesting cells, centrifugation helps pellet cells from the culture medium.
- Microcentrifuge tubes (0.5–2 mL): Commonly used in molecular biology workflows (DNA/RNA extraction, protein precipitation), where centrifugation concentrates or purifies samples.
- PCR tubes: Sometimes briefly centrifuged before amplification to collect droplets at the bottom and ensure proper mixing of reagents.

How to autoclave centrifuge tubes?
1. Check Material Compatibility
- PP centrifuge tubes and polycarbonate (PC) centrifuge test tubes: Most are autoclavable, but always check the manufacturer’s instructions.
- Polystyrene (PS) tubes: Not autoclavable – they will warp or melt.
- Glass centrifuge tubes: Autoclavable, but handle carefully to avoid breakage.
2. Prepare Tubes for Autoclaving
- Open the caps slightly (for screw-cap tubes) or leave them loose to allow steam penetration and prevent pressure buildup.
- Remove sealing films or non-autoclavable closures.
- Do not overfill: keep liquids at ≤2/3 of the tube volume to prevent boiling over.
3. Arrange Tubes
- Place tubes upright in an autoclavable rack or basket.
- If tubes are very small (e.g., microcentrifuge tubes), put them in an autoclavable tray with a lid to avoid scattering.
- Keep different plastics separate if possible (to prevent melting accidents).
4. Set Autoclave Conditions
- Standard sterilization cycle: 121 °C, 15 psi, 15–20 minutes.
- For larger volumes or resistant organisms: extend to 30–40 minutes.
- Use liquid cycle if tubes contain liquid (prevents rapid depressurization and boiling over).
5. After Autoclaving
- Allow pressure to normalize and temperature to drop before opening the door.
- Handle tubes with heat-resistant gloves.
- Tighten caps after cooling if needed for storage.
