Feature Of Vacuum Filtration Systems

  • Large volume processing capacity: designed for filtering 50mL to several liters of liquid, suitable for rapid processing of large batches of samples such as culture media, buffers, solvents, etc., significantly improving laboratory efficiency.
  • High throughput and automated operation: relying on vacuum filtration or pressurized drive, the pressure difference (such as a vacuum pump) pushes the liquid through the filter membrane quickly, with high throughput, suitable for continuous filtration, and reducing manual intervention.
  • Modular structure design: composed of filter head (including filter membrane), receiving bottle, and pressure interface, it is directly installed on the mouth of a standard reagent bottle, without the need to transfer liquid, reducing the risk of contamination.
  • The filter membrane material (such as PVDF, PTFE) and pore size (0.22μm/0.45μm/0.1μm) can be flexibly selected, compatible with aqueous phase, organic solvents and strong acids and alkalis.
  • Sterility and high cleanliness requirements: pre-sterilized filter heads (gamma ray or EO sterilization) can be directly used in sterile scenarios such as cell culture and injection preparation. The 0.22μm filter membrane can completely remove bacteria and ensure biosafety.
  • High chemical compatibility and durability: The filter membrane and sealing ring materials (such as PTFE resistant to organic solvents, PES resistant to high temperatures) adapt to a variety of samples and avoid swelling or corrosion.

FAQ of bottle top filtration system

What applications are bottletop filters commonly used for?

They are widely used for sterile filtration of cell culture media, buffer preparation, microbiological testing, pharmaceutical research, and laboratory sample clarification.

Vacuum filtration is more suitable for large-volume samples, provides higher throughput, and significantly reduces processing time compared with syringe filter.

Prepare the Equipment

  • Gather your bottle top filter unit (usually consisting of a filter cup, funnel, and receiver bottle), a vacuum source, and the liquid to be filtered.
  • Make sure all components are clean and sterile if required for biological or cell culture work.

Assemble the Bottle Top Filtration Unit

  • Attach the sterile bottle top filters securely to the receiver bottle (the collection bottle).
  • Ensure the connection is tight and leak-free.
  • Some bottle-top filters are screw-on types, while others use an adapter or quick connection.

Connect the Vacuum Line

  • Connect the vacuum tubing to the side arm or vacuum port on the vacuum filtration unit.
  • Make sure the tubing fits snugly to prevent air leakage.
  • Check that the vacuum pump is ready but not yet turned on.
  • Add the Liquid Sample
  • Carefully pour the liquid sample into the top funnel (filtration cup).
  • Do not exceed the maximum fill line marked on the cup to avoid overflow.

Start the Filtration

  • Turn on the vacuum pump gradually.
  • The negative pressure will draw the liquid through the membrane filter into the receiver bottle.
  • Filtration speed depends on the membrane type, pore size, and sample viscosity.

Complete and Disconnect

  • When all liquid has passed through, turn off the vacuum pump.
  • Carefully remove the vacuum tubing and unscrew the filter from the bottle.
  • Use sterile technique if the filtrate will be used for cell culture or sensitive analysis.

Storage or Disposal

  • The filtered liquid (filtrate) is now ready for use or storage.
  • Dispose of the filter unit appropriately if it is single-use (most bottle top vacuum filter are disposable).

1. Principle

Vacuum filtration operates on the same basic principle as gravity filtration, but it uses negative pressure (vacuum) to pull the liquid through the filter much faster.
The vacuum pump lowers the air pressure below the filter, creating a pressure difference that drives the liquid through the membrane or filter paper, leaving solids behind.

2. Components of a Vacuum Filtration System

A typical setup includes:

  • Vacuum pump or aspirator – provides suction force.
  • Filter funnel or bottle top filtration unit – holds the liquid and filter membrane.
  • Filter membrane – retains solid particles or microorganisms.
  • Receiving flask or bottle (filter flask) – collects the filtrate.
  • Rubber stopper or connector tubing – ensures an airtight seal between the funnel and flask.

3. Step-by-Step Process

  • Assemble the system: Connect the funnel to the receiving bottle or filter flask using an airtight adapter.
  • Add the filter membrane: Place a suitable membrane (e.g., bottle top filter 0.22 or 0.45 µm) in the funnel.
  • Pour the sample: Add the liquid to be filtered into the funnel.
  • Turn on the vacuum pump: The suction pulls the liquid through the membrane.
  • Collect the filtrate: The clear liquid passes through into the receiving bottle, while particles remain on the membrane.
  • Turn off the pump: Once filtration is complete, release the vacuum before disassembling the setup.
  • Nylon membranes offer strong mechanical strength and excellent chemical compatibility, making them suitable for both aqueous solutions and most organic solvents. Being hydrophilic, they offer fast filtration rates and minimal extractables, making them widely used in general laboratory filtration, HPLC sample preparation, and solvent purification. Due to their high compatibility, nylon membranes are versatile filtration membranes with a wide range of applications.
  • MCE membranes (mixed cellulose ester membranes) are naturally hydrophilic membranes with excellent flow rates and low protein adsorption. Their high biological cleanliness makes them suitable for microbiological analysis, such as bacterial filtration and collection, as well as for the filtration of various aqueous samples. MCE membranes perform particularly well when processing microbial, water, or environmental samples.
  • PES membrane (polyethersulfone) is one of the most commonly used filtration membranes in biological laboratories. It is a hydrophilic material with extremely fast filtration rates, extremely low protein adsorption, and strong chemical and thermal stability. PES membranes are ideal for sterile filtration of biological fluids such as cell culture media, serum, and buffers, making them a preferred material in cell and tissue culture.
  • CA membrane (cellulose acetate membrane) is also hydrophilic and characterized by extremely low protein adsorption, minimizing sample loss. It is particularly suitable for filtering biological samples containing proteins or enzymes. CA membranes effectively maintain the integrity of active ingredients when processing enzyme solutions, protein solutions, and various biological buffers.
  • PVDF (polyvinylidene fluoride) membranes are renowned for their excellent chemical resistance. Available in both hydrophilic and hydrophobic types, they offer excellent resistance to solvents, acids, and alkalis, as well as high mechanical strength. PVDF membranes are suitable for filtering samples containing organic solvents, corrosive liquids, or alcohols. They are also commonly used for gas sterilization filtration and organic solvent clarification.

Overall, PES membranes are most suitable for sterile filtration in cell culture applications; CA membranes are suitable for filtering biological samples requiring low protein adsorption; MCE membranes excel in microbiological analysis and water quality testing; Nylon membranes offer broad compatibility and are a general choice for general laboratory filtration; and PVDF membranes are best suited for filtering liquids containing organic solvents or those with high chemical aggressiveness.
