How to prepare microscope slides?
- Dry Mount (for dry, solid specimens): Used for: pollen, hair, feathers, plant fibers, insect parts, etc.
- Place the specimen directly on the center of a clean microscope slids.
- Carefully lower a cover glass (cover slip) on top to protect the sample and the lens.
- Observe under the microscope.
- Wet Mount (for living cells or transparent samples): Used for: pond water, blood, cheek cells, plant tissues.
- Place a thin microscopic glass slide or drop of the specimen in the center of the slide.
- Add 1–2 drops of water or another liquid medium (e.g., saline).
- Hold the cover slip at a 45° angle and slowly lower it to avoid trapping air bubbles.
- Blot away excess liquid with filter paper or tissue.
- Observe immediately.
- Stained Mount (to improve contrast and highlight structures): Used for: bacteria, onion epidermis, cheek cells, tissue samples.
- Prepare a wet mount as above.
- Add a drop of stain (commonly methylene blue, iodine, or safranin) at the edge of the cover slip.
- Place a piece of tissue paper on the opposite edge to draw the stain across the specimen by capillary action.
- Observe the enhanced contrast under the microscope.
How to clean microscope slides?
1. Initial Rinse
- Rinse the glass slide microscope under running distilled water to remove dust and large debris.
- Avoid tap water if possible, since minerals can leave residues.
2. Degreasing
- Wipe the microscope slider gently with 70% ethanol or isopropanol using lens paper or lint-free tissue.
This removes oils, fingerprints, and biological residues.
3. For Stubborn Contaminants
- Soak microscopic glass slides in a mild detergent solution or laboratory cleaning solution.
Rinse thoroughly with distilled water afterward.
4. Drying
- Air-dry slides in a dust-free area, or use compressed air.
If needed quickly, wipe gently with lens paper (never rough tissue, to avoid scratches).
5. Sterilization (for sensitive experiments)
- For microbiology or molecular biology applications, slides for microscope may need sterilization. Common methods include: Autoclaving (121 °C, 15 psi, 15–20 min); Dry heat sterilization (160–180 °C, 1–2 hours); UV sterilization (short exposure in a clean hood).
